Activated macrophages release a factor which lyses malignant cells but not normal cells

When rat macrophages are activated by exposure to bacterial endotoxin they acquire the capacity to lyse sarcoma cells in tissue culture. Although showing no immunological specificity this lytic effect was only detectable on malignant cells; normal cells were unaffected. This tumor-selective lytic effect is mediated by a labile supernatant product.

Hyaluronan receptors are expressed on human malignant mesothelioma cells but not on normal mesothelial cells.

Hyaluronan-binding sites were demonstrated on the cell surface of three malignant mesothelioma cell lines derived from human tumors using either [3H]hyaluronan or fluorescein-tagged hyaluronan. No hyaluronan-binding activity was observed on normal human mesothelial cells. The absence of hyaluronan receptors on normal human mesothelial cells was not due to a down-regulation by endogenously synth...

Activated Langerhans cells release tumor necrosis factor.

Langerhans cells act as antigen-presenting cells in immune reactions in the skin. What other roles they may play in inflammation is less well defined. We have tested whether these cells can produce TNF-alpha, an important mediator of inflammation. Resting Langerhans cells produce less than 0.1 U TNF-alpha/ml. Langerhans cells stimulated with phorbol myristate acetate (PMA) and lipopolysaccharid...

Normal cells, but not cancer cells, survive severe Plk1 depletion.

We previously reported the phenotype of depletion of polo-like kinase 1 (Plk1) using RNA interference (RNAi) and showed that p53 is stabilized in Plk1-depleted cancer cells. In this study, we further analyzed the Plk1 depletion-induced phenotype in both cancer cells and primary cells. The vector-based RNAi approach was used to evaluate the role of the p53 pathway in Plk1 depletion-induced apopt...

Histone deacetylase inhibitor induces DNA damage, which normal but not transformed cells can repair.

Histone deacetylase inhibitors (HDACi) developed as anti-cancer agents have a high degree of selectivity for killing cancer cells. HDACi induce acetylation of histones and nonhistone proteins, which affect gene expression, cell cycle progression, cell migration, and cell death. The mechanism of the tumor selective action of HDACi is unclear. Here, we show that the HDACi, vorinostat (Suberoylani...
